The GO term currently says 'protein binding' which is unacceptable. Since the protein has been shown to be involved in the stability of the extracellular matrix, the GO term GO:0005201 (extracellular matrix structural constituent) may be more appropriate.

The GO term contains a binding term, and therefore cannot be used. Also, the figure in question refers to a direct assay, so the evidence code should be changed to IDA.

It is unclear which figure is being annotated because the figure title is not denoted in the notes.

Change evidence code to IDA

The figure is of a direct assay and therefore the evidence code is incorrect

The GO:0097435 term is more appropriate for this because the data suggests that "the direct interaction between FN and Hsp90, as well as the decreased levels of both soluble and insoluble FN upon Hsp90 inhibition or knockdown, suggested that FN may be a new client protein for Hsp90 and that Hsp90 was involved in FN matrix assembly and/or stability." As shown in figure 7, inhibition of Hsp90 with NOV resulted in a reduction of extracellular FN matrix in Hs578T cells, The GO term 1990000 is not appropriate beacuse amyloid fibril formation was not looked at.

Evidence code should be from a direct assay.

The GO term contains a binding term, which is illegal to use. Instead, GO:1990000 Amyloid Fibril Formation may be appropriate. Fig. 5B shows treatment of Hsp90 increases the amount of insoluble FN. Fig. 6C shows treatment with KO or inhibited Hsp90 reduces insoluble FN. The authors state that the increase in insoluble FN suggests a role for Hsp90 in fibril formation.

The binding term problem was awarded points in February. No points for pointing it out again.

This annotation has been flagged because it has been edited since last assessment ||GO:0097435||fibril organization||PMID:24466266||IMP: Inferred from Mutant Phenotype||||P||An Interaction observed in vitro and colocalization of Hsp90 and FN in both normal and cancer cell lines were investigated using confocal microscopy and the DOC assay. These methods helped to determine the effect of exogenous Hsp90β on FN matrix formation and morphology in Hs578T cells. Treatment of the Hs578T cell line with exogenous soluble Hsp90β led to an increase in insoluble FN relative to soluble FN, suggesting a role for extracellular Hsp90 in fibril formation. Other methods used include co-immunoprecipitation, solid phase binding assay, immunoblot analysis, SPR spectroscopy and mass spectrometry.||complete
