Figure 4A shows that the protein Tfg3 is a component of TFIIF because it comigrates with components of TFIIF complex, and Figure 4B shows that when the complex is probed with anti-Tfg3 antibodies, there is a reaction showing that Tfg3 is present in the complex. Figure 4C-E shows that Tfg2 associates with common binding factors also part of the TFIIF complex, again by immunoprecipitation and antibody probing. Sequence homology does not prove that Tfg3 associates with the complex. Table 1 also shows via IMP/IDA that when tfg3 is deleted, there is significantly less transcription compared to wild type cells because the TFIIF complex cannot form correctly, and transcription is impacted. | 0 |
