To improve this annotation, I would suggest changing the notes to the following: Figure 4 shows that Ncs6p, a homolog previously found to affect TOR signaling in yeast, and ROL5 have similar functions. Figure 4a shows that ROL5 is involved in tRNA modification using a band shift assay. WT tRNAs exhibit a band shift in the presence of APM. This shift is absent from rold5-1 tRNA extracts. In the absence of APM there is no shift in either extract. Figure 4b shows that when the WT, the mutant ncs6 and the complemented mutant ncs6 are grown with rapamycin the mutant ncs6 presents retarded growth. Only by expressing ROL5 in the mutant ncs6 was the effect suppressed.

The assay does use mutants in comparison of WT to determine function in Fig 4b. The evidence code section states that IMP "trumps" IDA. I am not sure why the evidence code was marked as incorrect because I would still call this IMP.

The evidence code should instead be IDA.

The notes should instead state that: Figure 4B shows that when ncs6 is deleted, the growth of is inhibited in the presence of rapamycin and when rol5 is inserted (without the missense mutation) with the ncs6 deletion, the growth in the presence of rapamycin is not inhibited.
