Figure 5 shows the electrochemical assays performed on recombinant E. coli to determine the physiological effects of PRD1 endolysin (pMG118) involved in viral cytolysis. Host cell permeability (K+ and TPP+), cell growth and cell cytolysis (A550) were measured. Fig. 5a exhibited cell growth that steadily increased but never decreased, and variable changes in permeability. There was a peaked influx and efflux of K+, a decrease in TPP+, and steady, increased A550. The absence of endolysin and the presence of holin (pGZ9) failed to rupture cell membrane. Fig. 5e exhibited cytolysis with co-expressed holin and endolysin. Permeability and cell growth increased and peaked, but decreased dramatically below the initial baseline. The initial influx and latter efflux of K+, TPP+, and A550 values displayed how endolysin works to rupture host membrane when simultaneously present with holin in PRD1 for this synergistic effect.

@Kristina: Please read my annotation and the paper more thoroughly before you make a challenge. There are three measurements: K+ (permeability), TPP+ (permeability), and A550 (cell growth). Cell growth is probably the most important factor to consider in the figures listed. There is a constant growth of A550 but no decrease in Fig. 5a, signifying no cytolysis. Fig. 5e show an influx and efflux of all 3 measurements way below the baseline signifying cytolysis.

Kristina, there are no ECO IDs for biochemical assays because it's synonymous to other evidence ontologies. If you actually read the article, the experiment specifically lists Electrochemical assays that were conducted to determine E.coli cell physiology. Specific terms are preferred over broader categories. With that said, ECO:0006006 electrophysiology assay evidence used in manual assertion is the best description of evidence for this figure describing endolysin. A similar annotation for holin was also created and approved.

I think you need an IDA (Inferred from Direct Assay) experimental evidence code attached to this annotation. The authors are conducting a biochemical assay (i.e. measuring K+ leakage and ΔΨ) using recombinant E. coli in order to measure the normal function of the pMG118 gene product. This fits with the IDA evidence code.
