Mutant strains of sindbis virus with insertion sequences in the PE2 glycoprotein gene have significantly reduced binding capabilities. PE2 cleavage is necessary for E2/E3 specific spike protein with E2 specific for receptor binding.

Table 1 represents the results of the mutations on various clones to express MTase activity. Changes and/or deletions to conserved amino acid sequences in nsP1 resulted in lower Km for S-adenosyl methionine. Reduced methyl donor substrate concentration significantly reduced MTase activity and viral infectivity.
