This aldolase activity of proA is correct, however, this is the case in Pseudamonas Ochraceae, not in E.coli. E.coli was simply used as a vector for overexpressing proA. 2. Figure 5(B) is the best figure that shows that the gene product of proA and the HMG aldolase enzyme are identical. However, this is not done by expression level, but by the molecular weight of the proteins. abouSDS-PAGE measures molecular weight. 3. It's also important to mention Table 3! This shows empirical evidence about the kinetic constants of the proA gene product (which they cite to be similar to those of the HMG aldolase of P.ochraceae in other papers where they have done studies on the isolated protein.) | 8 |
