Abstract |
The bacteriophage SPO1 DNA polymerase-encoding gene, which contains a self-splicing intron, has been sequenced and its amino acid (aa) sequence has been deduced. The aa sequence of SPO1 DNA polymerase shows a high degree of similarity with that of DNA polymerase I from Escherichia coli (Po1I). Alignment with the sequences of Po1I, and the phi 29 and SPO1 DNA polymerases indicate that the aa residues that have been implicated in 3'----5' exonuclease activities are conserved. |

Matched 100% with Klenow fragment of DNA with an E-value of 3E-105. Figure 2 linked in the PubMed article shows both the nt and aa sequences of the DNA polymerase gene. These sequences were put together through experiments to assemble gene 31. When the subclones generated from the experiment were deemed too long and the resolution was poor, synthetic oligodeoxynucleotides were used as primers. Standard methods were used to manipulate the DNA. From the figure, it is said that "sequence data will appear in the GenBank Nuclcotide Sequence Database under accession No. M84415." |
