In the paper, the gene sequence of Diaphorina citri is compared in a sequence analysis with Nilaparvata lugens, Aphis citricidus, Riportus pedestris, and Acyrthosiphon pisum.

Figure 1 shows this sequence alignment. The other bacteria in the alignment display similar cathepsin B-like cysteine peptidase activity from hemipterans. The dashed boxes in Figure 1 display a probable occlusion loop characteristic of cathepsin B-like cysteine peptidases.

Experimenters conducted a sequence alignment with Liberibacter asiaticus (strain 62) with Escherichia coli (strain K-12) to confirm that serine-type pepsidase I (SPase I) is conserved. The sequence alignment was also comparing Liberibacter asiaticus (strain:gxpsy) and Liberibacter asiaticus (strain:Ishi-1).

Suplementary Figure 1 shows the identity for the SPase I of the three Liberibacter asiaticus strains is 100%. The Liberibacter asiaticus SPase I protein shares 34% identity and 52% similarity with that of E. coli.

Liberibacter asiaticus lepB could partially complement the lepB mutant of E.coli. lepB is the gene that encodes Serine peptidase I.

Figure 4 shows that the wild type E. coli grew at 37 degrees celcius and 42. The amber E. coli mutant grew at 37 but not 42. The lepB homolog of Liberibacter asiaticus partially restores growth of the mutant strain to result in growth at 37 and 42.
