I would change the notes to explain Figures 4A and 4B as well since these describe how the HERC2 gene and OCA2 expression are related by HLFT binding.

Figure 4A: ChIP-qPCR found that HLFT has better binding with the T-allele of HERC2 r12913832 (found in HEMn-DP cells) than the C-allele found in HEMn-LP cells. Figure 4B: Overexpression of HLFT caused increased OCA2 expression only in HEMn-DP cells. The increased OCA2 expression leads to increased pigmentation.

The notes are not very clear on how exactly this provides proof for this protein. I have made new notes with a different figure.

Notes: The HERC2 enhancer is controlled by several transcription factors. Figure 4A-D shows ChIP-qPCR data of the mRNA expression of the different enhancers. Human melanocytes with a SNP rs12913832 of a C to T-allele are more darkly pigmented. Cells with the C-allele and Cells with the T-allele SNP were compared and mRNA expression of the enhancers was measured. The T-allele SNP having cells produced significantly larger amounts of mRNA expression for the enhancers suggesting that this protein plays a role pigmentation.

Note that there are other accepted annotations to this paper. This paper is not about the function of the HERC2 gene. It shows that a SNP in the HERC2 gene creates an enhancer that regulates expression of OCA2. Regulation of transcription from that enhancer is controlled by other proteins.
