Deficiency in SESN2 is a mutant phenotype (difference between alleles), so the evidence code should be IMP (which trumps IDA). Also, the data given in Figure 1 indirectly links Sestrin2 to AMPK. Sestrins are stress-inducible proteins that activate AMPK, and AMPK activation reverses glucose intolerance and insulin resistance. Deficiency in Sestrin2 aggravated glucose intolerance and insulin resistance (due to leptin deficiency). The GO term should be regulation of protein phosphorylation (GO:0001932), as evidenced by Figure 3. Sestrin2 is shown to regulate AMPK signaling, and the evidence code should be IMP. | 4 |
