Challenge | Sejal, Team UCL-Maple Leafs | 2011-10-16 13:13:47 CDT | I understand your reason, but figure 1A shows immunostaining of cultured cortical neurons of endogenous IL1RAPL1 and antibodies against postsynaptic marker (MAP2 and Shank1) and presynaptic marker (Bassoon). Therefore the overlap in the staining of IL1RAPL1 with Shank1 indicating it is located on the dendrite. We still cannot be positive that figure 1A show synapse assembly because other genes can be involved in synapse assembly that have not be stained for. The only way to find out is knocking out IL1RAPL1, which is seen in figure 1C. Figure 1C can be annotated to synapse assembly because in this figure IL1RAPL1 is knocked out by siRNA and we can see a decrease in Bassoon marker which is a presynaptic membrane marker, which is why I annotated figure 1C to Presynaptic membrane assembly (this a child term for synapse assembly) | 0 |
