Figure 6: E. coli transformed with pUC19-TRM1 show tRNA M(2,2)guanine-methyltransferase activity, while neither E. coli transformed with an empty plasmid nor wild type show that activity. IMP.

Sorry for separating these all into separate challenges. The strain is ATCC 204508 / S288c.

Also, Uniprot calls the protein "tRNA (guanine(26)-N(2))-dimethyltransferase, mitochondrial." The paper simply calls the protein TRM1.

Based on Figure 6, the TRM1 gene codes for the structural proteins that causes tRNA M(2,2) guanine- methyltransferase activity, as expressed in the deficient cells seen with the open circle plot in the E. coli cells that were targeted for TRM1 expression.

Notes have incorrect organism. Organism should be: Saccharomyces cerevisiae (strain ATCC 204508 / S288c) (Baker's yeast).

Challenging clarification in the notes portion. Figure 6 shows the TRM1 gene restores the m2/2guanosinemethyltransferase activity in deficient yeast cells.

Figure 6 shows that pUC19 – TRM1 transformed yeast cells: m2/2guanosine-methyltransferase -deficient tRNA incorporated methyl in large amounts while the wildtype tRNA incorporated low amounts of methyl. The pUC19 transformed cells wildtype tRNA and m2/2G-deficient tRNA incorporated low amounts of methyl as well. Showing that TRM1 gene catalyzes modification of the tRNA m2/2Gmethyltransferase of m2/2G-deficient yeast tRNA.

1 point for providing better notes, but they do not fully fix the problem.
