  • Protein: Q8L1D1
  • E. faecalis OG1RF with conjugative plasmid pIP501 with deletion of TraG protein had no detectable transconjugants, while bacteria expressing the functional TraG protein had transconjugants within the expected range, indicating TraG is necessary for conjugation. “Assuming the indispensability of TraG, no transfer should occur in the traG-knockout mutant in a biparental mating in G bacteria, whereas transfer rates with an isogenic, traG-proficient pIP501 plasmid should be in the expected range of approximately 105 transconjugants per recipient.” … “Transfer rates of the traG deletion mutant were below the detection limit of the assay (<3.5X10^-9+/-2.4X10^-9, mean value of three independent assays), whereas transfer rates with the isogenic strain E. faecalis OG1RF(pIP501) were at least 20,000 times higher and in the expected range for pIP501 of 7.6X10^-5+/-5.7X10^-5 transconjugants per recipient cell (mean value of three independent assays).” Data not shown. IMP.

The current submission is missing an evidence code. The applied evidence code should be IMP because a deletion is a mutation of the genome to observe the effects in absentia of the TraG.

GO:0009291 unidirectional conjugation PMID:23913323 IMP: Inferred from Mutant Phenotype P Figure 2.

Immunoblot assay with protein lysates obtained from E. faecalis OG1RF (pIP501) and E. faecalis OG1RF (pIP501ΔtraG) to show the expression levels of traH (image A) , traK (image B) , and traM (image C) were unaffected by the traG-knockout mutant. Proving that when roughly 97% of the traG coding region was deleted, there was no negative effect on expression of the downstream genes in the pIP501 tra operon. ||complete
