I think that portal proteins are more structural while sequence similarity is referring to the similarity in base pairs. The notes do not explain the connection between a genetic sequence and the cellular component/ structure of the protein.

Does not say how they experimentally determined that its sequence and protein is similar enough to another portal protein to confirm that it is indeed a portal protein in your notes.
