Figure 8A-B. All three potential forms of TcdE (tcdEM1) or only M25 and M27 (tcdEM25) or only M27 (tcdEM27) were used. When expressed in E. coli, the TcdEM1 (pCD311) and TcdEM25 (pCD302) forms induced complete bacterial lysis within five minutes after thermo-induction whereas expression of the TcdEM27 form (pCD322) caused complete lysis only at about 40 min. As was the case for λS protein, lysis induced by expression of any of the putative forms of TcdE required expression of the λ endolysin. However, when expressed in E. coli λCmrΔ(SR), carrying a deletion in holin and endolysin genes, the λS, TcdEM1, TcdME25 and TcdEM27 forms did not cause lysis, indicating that the lysis in E. coli could not be due to over-expression of the λS and TcdE forms. |

Figure 3A and 3B. In Fig 3A, there was a large reduction in the amount of toxin secreted by the mutant when compared to the wild-type strain. In Fig 3B, there is more toxin accumulated in the cytoplasm of the tcdE mutant than in the wild-type strain in stationary phase cells. |
