HeLa cells express Ataxin-7 endogenously. In figure 5, HeLa cell strains expressing either wild type and mutant Ataxin-7 protein were treated with noncodazole, an inhibitor of cytoskeletal microtubule polymerization. Cells with wild type Ataxin-7 maintained the normal filamentous pattern of cytoskeletal microtubule organization, while Ataxin-7 mutants showed loss of the filamentous microtubule organization.


Figure 6 shows that HeLa cells with Ataxin-7 gene silenced by siRNA are unable to maintain a filamentous microtubule network in the presence of noncodazole. Furthermore, HeLa cells with silenced Ataxin-7 gene by siRNA were also unable to recover their microtubule networks. Altogether, these results show that Ataxin-7 prevents (negatively regulates) microtubule depolymerization, hence stabilizing microtubules.
