Annotation to GO:0004396 ! hexokinase activity, GO:0004340 ! glucokinase activity, GO:0008865 ! fructokinase activity, etc in the GO sense is not the same as applying the name of an enzyme. Enzymes named Hexokinase and Glucokinase are distinct in their substrate specificities. Although Enzymes in the true glucokinase family do not phosphorylate other sugars., annotating to Glucokinase activity only asserts that the enzyme can phosphorylate glucose, not that it does not phosphorylate other hexose sugars.
In GO annotation substrate specificity should be captured by using NOT annotations for substrates that are not used at physiological concentrations.
