In part A, there are two final products by cleaving a plasmid pIR9 with Holliday junction resolvase; nicked circular or linearized plasmid. Nicked circular plasmid is obtained by only one incision done in the DNA while linearized plasmid is done by two incisions. In part B, it shows an endonuclease assay for AtGEN1 and AtSEND1 in order to observe if both protein have a similar function in the organism. The uncleaved supercoiled plasmid was separated to have the nicked circular and the linearized plasmid product. Then, the endonuclease assay shows that the nicked circular plasmid is the slowest to run in the gel because it is found almost on the top. Also, it indicates that both proteins activity actually are the same using negative controls D75A for AtGEN1 and D76A for AtSEND1. The graph also points out that both resolvases proteins act similar in which AtGEN1 maximizes after 20 minutes and AtSEND1 in 60 minutes. complete
