Figure 2. Experiment performed in which cultures were analyzed by immunoblotting antibody-specific for hNIFK. Proteins that interacted with Ki-FHA were pulled down with a GST-Ki-FHA fusion protein and subsequently analyzed with Western blot with the anti-NIFK antibodies. As shown in Fig. 2 A, the mitotic forms of hNIFK (bands I and II) were pulled down a sufficient amount to conclude that hNIFK interacts efficiently with Ki-HFA binding domain.

Figure 5. Immunoblotting experiment conducted to see whether hNIFK binding to pKi-67 is dependent on mitotic phosphorylation of hNIFK. Interacting proteins had bars that were pulled down n immunoblot. NIFK was pulled down efficiently (Fig. 5A) T234A and T238A mutants of hNIFK were pulled down less efficiently - proving mitotic phosphorylation is a necessary component of binding between hNIFK and pi-67.
