E. coli has a single CPS (carbamoyl phosphate synthetase) encoded by 2 genes, carA and carB. CPS is comprised of a heterodimer of CarA (referred to as the small subunit) and CarB (large subunit). CarA is responsible for binding of glutamine at a conserved catalytic dyad (Cys-His) and initiating the hydrolysis of glutamine to glutamate and ammonia. CarA then transfer of the ammonia group through an ammonia tunnel to the CarB subunit. The large subunit (CarB) then catalyzes an initial round of phosphorylation of bicarbonate to carbamate at the first active site. This product of this reaction travels down the carbamate tunnel where a second phosphorylation reaction of carbamate produces carbamoyl phosphate at the second active site in CarB. CarB is not capable of producing carbamoyl phosphate without CarA in the presence of glutamine. However, CarB can catalyze its production in the presence of ammonia as the nitrogen donor. The next two steps in the synthesis of pyrimidine ribonucleotides are catalyzed by 2 separate enzymes: aspartate transcarbamoylase and dihydroorotase.[1]
Interestingly, higher eukaryotes have multiple CPS enzymes. CAD is a multifunctional protein that encodes CPS function, as well as aspartate transcarbamoylase and dihydroorotase.[1]
