Comparison of the pattern of fluorescence from GFP–ZapA with that from the membrane dye TMA-DPH showed that transverse bands of the fusion protein were often present at the midcell position in cells in which no septum could be detected. This finding is consistent with the idea that ZapA localization precedes septum formation, as is true for FtsZ (Fig. 2A).
