Hydrophobic interaction allowed the removal of E. coli SecA eluting at higher ammonium sulphate concentration than the B. subtilis SecA (not shown). Superdex 200 gel filtration was employed to remove minor impurities detected on stained gels, yielding a homogenous SecA protein (98-99% pure on the basis of Coomassie brilliant blue staining) which migrates near to its predicted molecular mass of 95 kDa on SDS-PAGE (Fig. 1, lane 3). The purified protein reacted weakly with antiserum raised against the E. coli SecA(lane 4), while an intense band was observed with antiserum directed against purified B. subtilis SecA protein(lane 5).
